Modern protein purification methods use small particles, high plate-count resins that require both high pressure, precise flow rates and precise gradients to achieve the highest purity of the final product thus eliminating any off-target results (artifacts). These proteins will be used for the study of reactions of nanobodies, receptor structural biology, and drug development. Specific requirements: The AKTA Go purification system with UNICORN software must be very versatile and capable of purifying proteins with high hydrophobicity. Titanium pump heads help to avoid the buildup of hydrophobic products within the pump head. Further, this device must avoid the use of PEEK plastic as much as possible, as PEEK is hydrophobic and can cause protein buildup. Comparable product lines use a hot lamp for flow cell readings causing temperature changes and repeated lamp changes. This unit instead uses a LED system to avoid these issues. The unit must be able to workflow rates up to 25 mL/m and with a pressure up to 5 MPa to ensure the ability to run small particle high plate-count columns. A fraction collector is needed to isolate the portions pure product. In addition, the UNICORN software is needed for continuity with respect to previous research and to ensure the previous developed methods will perform as expected. The AKTA Go purification system with UNICORN software is assembled based on the configuration.
